Children?s body fatness linked to decisions made in the womb

ScienceDaily (Aug. 22, 2012) ? New born human infants have the largest brains among primates, but also the highest proportion of body fat. Before birth, if the supply of nutrients from the mother through the placenta is limited or unbalanced, the developing baby faces a dilemma: should resources be allocated to brain growth, or to fat deposition for use as an energy reserve during the early months after birth?

Scientists at the University of Southampton have shown that this decision could have an effect on how fat we are as children.

In new research, published in the journal PLoS ONE on August 22, 2012), scientists at the Medical Research Council (MRC) Lifecourse Epidemiology Unit at the University, performed ultrasound scans on 381 pregnant women taking part in the Southampton Women's Survey. They measured the blood flow from the placenta to the unborn baby, and the distribution of this blood to either the liver of the baby or bypassing the liver to supply the brain and heart. This was then compared with the infant's body fatness at birth and at four years old.

The findings show that greater blood flow to the baby's liver in late pregnancy was associated with greater body fatness in the infant at birth and at age four. In contrast, lower liver blood flow and a "brain-sparing" blood flow pattern (when the blood bypasses the liver and goes to the brain) occurred when the placenta was smaller and less able to meet baby's demand for essential nutrients in the womb.

These findings were independent of an association between mother's body fatness and the body fatness of her infant.

Keith Godfrey, Professor of Epidemiology and Human Development, at the University who led the study, explains: "In our evolutionary past, the demands of a big brain have led the unborn baby to develop blood flow responses which preserve nutrient delivery to the brain when the supply of essential nutrients from the mother cannot meet the baby's requirements.

"However, having a big brain has also led to evolution of a strategy to adjust blood flow through the baby's liver, which enables the liver to produce more fat -- this acts as an energy reserve, protecting brain development during periods of illness or under-nutrition in early infancy. Our data suggests that evolution of this strategy has brought with it a predisposition to obesity and later diabetes in contemporary societies with abundant nutrition in later postnatal life."

Professors Guttorm Haugen from the University of Oslo and Torvid Kiserud from the University of Bergen were part of the research team. They comment: "An interpretation of our findings is that there could be programmed effects on the liver that arise from blood flow adaptations in the womb and predispose individuals to gain excess body fat. Although further studies are needed, our findings add weight to current concerns that the current epidemic of childhood obesity and associated disorders may partly have its origins through adaptations made by the developing baby during pregnancy."

Professor Mark Hanson, Director of the University of Southampton's Human Development and Health Academic Unit, adds: "If the supply of nutrients across the placenta is inadequate or unbalanced, the unborn baby has to decide whether to prioritise fat deposition or spare brain growth -- it does this by changing the amount of blood flowing to the liver and brain. A decision to increase blood flow to the liver has lasting implications for the child's body fatness.

"Transfer processes across the placenta for some nutrients such as glucose evolved in environments less affluent than those now prevalent in developed populations, and our findings additionally suggest that in circumstances of maternal obesity and nutrient excess these processes now also lead to excessive fat deposition in the womb."

"This strengthens the case for all women of reproductive age having greater access to nutritional, education and lifestyle support to reduce the risk of obesity in their children and improve the health of the next generation."

Professor Cyrus Cooper, Director of the MRC Lifecourse Epidemiology Unit comments: "This study is part of a wider body of work by the MRC Lifecourse Epidemiology Unit into how factors during pregnancy might have a long-term influence on childhood growth and development. This is a wonderful example of multi-disciplinary research using the unique clinical resource provided by the Southampton Women's Survey."

Helium-breathing apes display operatic vocal skills, say researchers

The gibbon, a lesser ape native to Asia, uses the same vocal techniques as trained sopranos, finds a Japanese experiment involving helium-enriched air.

The Japanese study provides evidence for an unusual physiological similarity between gibbons and humans.

"The complexity of human speech is unique among primates as it requires varied soft sounds made by the rapid movements of vocal tracts," lead researcher Takeshi Nishimura, from the Primate Research Institute at Kyoto University, said in a statement. "Our speech was thought to have evolved through specific modifications in our vocal anatomy. However, we've shown how the gibbons' distinctive song uses the same vocal mechanics as soprano singers, revealing a fundamental similarity with humans."

Nishimura's team analyzed 20 melodious and loud calls of a captive white-handed gibbon (Hylobates lar) in a normal atmosphere, followed by 37 calls in an environment infused with helium.

Sucking in helium from a balloon gives humans a squeaky, high-pitch-sounding voice, because the gas, which is less dense than air, pushes the resonance frequencies of the vocal tract upwards, but doesn't change the sound at its source. An analysis of the gibbon's squeaky songs suggested that the same is true for these apes. Like humans, the origin of the sound of a gibbon's call, which occurs in the larynx, is separate from the vocal tools used to modify it, the research showed. [Listen to Gibbon Calls]

What's more, the analysis demonstrated gibbons have expert control over the tuning of their vocal cords and tract when singing ? an ability that is important to the subtleties of human speech and is mastered by soprano singers.

"This is the first evidence that gibbons always sing using soprano techniques, a difficult [vocalization] ability for humans which is only mastered by professional opera singers," Nishimura said. "This gives us a new appreciation of the evolution of speech in gibbons while revealing that the physiological foundation in human speech is not so unique."

The study was published this week in the American Journal of Physical Anthropology.

Australian cricketers will handle UAE weather: coach

Australia's acting coach Steve Rixon on Wednesday hoped his players will adjust to the hot conditions in United Arab Emirates during their matches against Afghanistan and Pakistan.

Australia's squad arrived here Wednesday with the temperature as high as 43-degrees Celsius, but Rixon hopes sleeping late and breakfast at noon will help during the series in which one-day games will start as late as 6pm in the evening.

Australia take on Afghanistan in the first-ever one-day game between the two countries in Sharjah on Saturday before facing Pakistan in three one-day and as many Twenty20 games.

The pre-series hype focused on how Australia will cope up with the high temperatures but Rixon said a policy has been devised to combat the oppressive weather.

"Our guys will have to eat late in night and just start the pattern. We need to start the patterns now so by the time the first game comes around, we'll be starting to build some sort of body clock that's going to work for us," Rixon told a press conference.

The 58-year-old former Australian wicket-keeper batsman is standing in for head coach Mickey Arthur, who will join the tour later, having been busy working plans for next month's World Twenty20.

Rixon said the team had prepared to combat heat.

"The weather was very warm up in Darwin where we camped but nothing as oppressive as over here. We're well aware of how hot it's going to be. If we sit here and talk about the weather all day, we're not getting the job done.

"We don't see the sun," he said. "We'll be in night-cricket mode, although it will be warm. But we're not getting the (sun) rebound off the ground which is always a big problems at most cricket grounds.

"So to me, just get on with it."

Rixon said the matches in UAE will help the team tune up for World Twenty20 in Sri Lanka.

"Whatever happens here will be a very big stepping stone to us being successful in Sri Lanka," said Rixon of Australia who slipped to fourth in one-day rankings for the first time in three years.

Rixon said players are now accustomed of playing in heat.

"I think a lot of players in the modern day games playing a lot more in the sub-continent with IPL (Indian Premier League) and other forms of cricket played in the sub continent in place like Chennai where it is very warm."

The acting coach agreed Afghanistan will be an unknown quantity, but was sure his team can beat them.

"Afghanistan has come in as a minor contender but they are now competiting at the top and best level. So we have got to come in with every answer and respect for the opposition.

"We also like to think that we are going with the upper hand to be able to beat them in these sort of conditions with the sort of side we have with us."

Rixon said Australia have prepared well to tackle Pakistan's much vaunted spin-attack, led by Saeed Ajmal.

"The emphasis on Darwin camp was on spin. So we are looking at the strength of the opposition. We are trying to play to our strengths more importantly but we have prepared well for Pakistan," said Rixon.

Pakistan and Australia will play the first one-day in Sharjah on August 28 followed by second in Abu Dhabi (August 31) and Sharjah (September 3). All three T20 matches will be played in Dubai on September 5, 7 and 10.

The code determines the recognition of RNA molecules by a superfamily of RNA-binding proteins called pentatricopeptide repeat (PPR) proteins.

When a gene is switched on, it is copied into RNA. This RNA is then used to make proteins that are required by the organism for all of its vital functions. If a gene is defective, its RNA copy and the proteins made from this will also be defective. This forms the basis of many terrible genetic disorders in humans.

RNA-binding PPR proteins could revolutionise the way we treat disease. Their secret is their versatility - they can find and bind a specific RNA molecule, and have the capacity to correct it if it is defective, or destroy it if it is detrimental. They can also help ramp up production of proteins required for growth and development.

The new paper in PLOS Genetics describes for the first time how PPR proteins recognise their RNA targets via an easy-to-understand code. This mechanism mimics the simplicity and predictability of the pairing between DNA strands described by Watson and Crick 60 years ago, but at a protein/RNA interface.

This exceptional breakthrough comes from an international, interdisciplinary research team including UWA researchers Professor Ian Small and Aaron Yap from the ARC Centre for Excellence in Plant Energy Biology and Professor Charlie Bond and Yee Seng Chong from UWA's School of Chemistry and Biochemistry, along with Professor Alice Barkan's team at the University of Oregon. This research was publicly funded by the ARC and the WA State Government in Australia and the NSF in the USA.

"Many PPR proteins are vitally important, but we don't know what they do. Now we've cracked the code, we can find out," said ARC Plant Energy Biology Director Ian Small.

"What's more, we can now design our own synthetic proteins to target any RNA sequence we choose - this should allow us to control the expression of genes in new ways that just weren't available before. The potential is really exciting."

"This discovery was made in plants but is applicable across many species as PPR proteins are found in humans and animals too," says Professor Bond.
